In this work, we use NMR and MD simulations to investigate the molecular mechanisms underlying activation of the human RNA demethylase Alkbh5. We show that binding of the methylated substrate induces a metal-centered conformational rearrangement that activates catalysis and confers substrate selectivity to Alkbh5. In this work, we characterize the link between the kinetics of the of the g+-to-g- rotameric transition of bacterial Enzyme I and its enzymatic activity. In addition, we describe how to engineer hybrid mesophilic/thermophilic chimeras that assist investigations of the dynamics/function relationship in proteins.
